В настоящей работе мы оценили, может ли прекондиционирование, заключающееся в легком механическом повреждении ногтевого ложа, сместить ответ тканей в сторону регенерации. У мышей C57/BL6 с помощью иглодержателя или микрометра выполнялось сдавление ногтевого ложа, после чего проводили субкритическую ампутацию кончиков пальцев по косой линии через определенные временные интервалы (1 час, 1 день, 3 дня или 5 дней) после сдавления. Мы оценили динамику внешней морфологии и выполнили гистологическое исследование на 42-й день после ампутации, после чего интерпретировали результаты с использованием разработанной нами шкалы балльной оценки. Прекондиционирование значимо улучшало частоту и качество частично регенеративных исходов: высокая степень анатомического восстановления (3 и 4 балла) наблюдалась преимущественно в группах с прекондиционированием, а качество исхода коррелировало с ранним появлением ногтя после его ампутации. Эффекты прекондиционирования не зависели от временного интервала между сдавлением и ампутацией, однако для них была характерна выраженная поляризация в зависимости от силы воздействия. Умеренная компрессия (25 и 50% толщины ногтя) усиливала регенерацию, тогда как чрезмерная (75%) — приводила только к нерегенеративным исходам. Гистологический анализ показал, что частично регенеративные исходы сочетались с устойчивым сохранением близкого расположения остаточных костных структур дистальной фаланги (P3) и эпителия ногтевой матрицы. Наши данные свидетельствуют о том, что прекондиционирование путем сдавления ногтевого ложа смещает баланс от нерегенеративного заживления в сторону эпиморфной регенерации. Полученные результаты потенциально обосновывают подход с прорегенеративной подготовкой тканей перед хирургическими вмешательствами.</p></abstract><trans-abstract xml:lang="en"><p>Stimulation of epimorphic digit tip regeneration under non-regenerative conditions remains a pivotal goal in regenerative biology and medicine. Our study investigates whether tissue preconditioning by mild injury can shift the response toward regenerative outcome. In adult male C57/BL6 mice mechanical occlusion was applied to the nail bed using either a needle holder or a micrometer followed by subcritical oblique digit tip amputation at specific time intervals (1 hour, 1 day, 3 days, or 5 days). We analyzed dynamic gross morphology and histology at 42 days post-amputation and then assessed results using an in-house developed scoring scale. Pre-conditioning significantly improved the frequency and quality of partially regenerative outcomes in the used model. High-grade anatomical restoration (scores 3 and 4) appeared predominantly in the pre-conditioned groups and the quality of outcome correlated with early nail appearance. The effect of pre-conditioning was independent of the time interval between occlusion and amputation. However, it exhibited a strict biphasic, strength-dependent profile: moderate compression (25% and 50%) enhanced regrowth, while excessive compression (75%) completely abolished regeneration. Histological analysis revealed that partially regenerative outcomes were consistently coupled with sustained spatial proximity between the residual distal phalanx (P3) bone structures and the nail matrix epithelium. Our data indicates that pre-conditioning by nail bed occlusion shifts the balance from non-regenerative healing toward epimorphic regeneration.
